Connection to Box/Pc

in Samsung Pro Tool it shows me my device under COM and ADB, when I connect the phone with USB cable to the PC.

does the phone need to be connected to the RJ45 connector for flashing?

Depends on phone. If you can't switch to usb you have to use RJ45 cable
